Background
eIF2α (eukaryotic initiation factor 2 subunit alpha) is a 35 kDa phosphoprotein that, together with the G-protein βγ dimer eIF2B, forms the heterotrimeric eIF2 complex essential for delivering the initiator methionyl-tRNA to the 40S ribosomal subunit in a GTP-dependent manner; phosphorylation of Ser51 by stress-sensing kinases (PKR, PERK, HRI, GCN2) converts eIF2 into a non-competitive inhibitor of its guanine-nucleotide-exchange factor eIF2B, thereby repressing global cap-dependent translation while selectively permitting ATF4 and other stress-response mRNAs, a switch that underlies cellular adaptation to starvation, oxidative stress, viral infection, and proteotoxicity, and whose dysregulation contributes to diseases ranging from neurodegeneration and cancer to diabetes and viral pathogenesis.
